Microcrystalline cellulose (MCC) has been widely regarded as an indispensable excipient for Extrusion/Spheronization. Various excipients like crosslinked polyvinyl pyrrolidone, HPMC, PEG, Gelucire, methyl cellulose, cellulose acetate, carrageenan, starch and sodium alginate were screened for their function as spheronization aids.

Various excipients like crosslinked polyvinyl pyrrolidone, HPMC, PEG, Gelucire, methyl cellulose, cellulose acetate, carrageenan, starch and sodium alginate were screened for their function as spheronization aids. Placebo pellets of the selected alternative spheronization aids were made and compared with MCC in terms of formulation aspects, process parameters and physical properties of the pellets. An attempt was made to incorporate the selected alternative spheronizing aids into three models drugs and study the effect of drug solubility, size, morphology and loading on process parameters and physical properties of the pellets and was compared with MCC.
