Introduction
It is a process by which a drug leaves a drug product and is subjected to ADME and eventually becoming available for pharmacological action.
It involves the study of drug release rate, dissolution/ diffusion/ erosion studies .
Drug release kinetics is application of mathematical models to drug release process.
Drug release mechanism and kinetics are the two important characteristics of a delivery system in describing the drug dissolution profile.
Kinetic Models
The kinetic models are used to evaluate the kinetics and mechanism of drug release from the tablets.
The model that best fits the release data is selected based on the correlation coefficient ® value in various models.
The model that gives high ‘r’ value is considered as the best fit of the release data.
The “father” of mathematical modeling of drug delivery is Professor Takeru Higuchi.
Exploratory Data Analysis methods
Exploratory data analysis methods are the method is useful in obtaining an improved understanding of the dissolution data of controlled release formulation.
This method can be used in the first step to compare dissolution profile data in both graphical and numerical manner.
Then, the data of the dissolution profiles are summarized numerically and 95% confidence intervals for the differences in the mean dissolution profiles at each dissolution time point are evaluated.
The dissolution profile data are illustrated graphically by plotting the mean dissolution profile data for each formulation with error bars extending to two standard errors at each dissolution time point.
Conclusion
Drug release kinetics helps efficiently in in vitro study of drug release and kinetic modeling predicts relationship between drug dissolution and geometry on the drug release patterns mathematically .
